The ChampionChIP Modified Histone Antibody Kits provide the specific antibodies and real-time PCR positive and negative control primers needed for a complete and reliable analysis of the histone modifications that regulate chromatin structure and accessibility. Its modified histone-specific ChIP-grade antibody has been rigorously validated for efficient and specific enrichment of DNA representative of either transcriptionally active euchromatin, or transcriptionally inactive euchromatin, or heterochromatin using a panel of real-time PCR primers. The matched control IgG provided in the kit has also been extensively tested for minimal co-immunoprecipitation of genomic DNA. The control qPCR primers detecting DNA representative of different chromatin states are included for the appropriate positive and negative controls to insure the efficiency and specificity of each ChIP sample preparation. How It Works: Speed and Ease

Immunoprecipitation Done in 2 Hours with ChampionChIP Antibody Using an optimized IP buffer system provided by the ChampionChIP One-Day Kit, the efficiency and specificity of the ChIP-grade antibody are maximized such that immunoprecicipation can be done within 2 hours. Sonicated chromatin from HeLa cells (20 µg) was immunoprecipitated with 2 µg of anti-H3K4me2 antibody (blue bars) or control IgG (red bars) for 2 h using the ChampionChIP One-Day Kit (GA-101). The resulting input and ChIP DNA fractions were characterized with qPCR primers specific for the GAPDH, MYOD1 and SAT2 genomic loci to calculate the percentage of co-precipitating DNA relative to input. The immunoprecipitation using anti-H3K4me2 enriched genomic DNA representing actively transcribed euchromatin (the GAPDH locus) with a high signal-to-noise ratio, demonstrating the efficiency of the Antibody and One-Day ChIP Kit combination. In contrast, the immunoprecipitation did not significantly enrich genomic DNA representing either transciptionally inactive euchromatin (the MYOD1 locus) or heterochromatin (the SAT2 locus) relative to the immunoprecipitation using control IgG, demonstrating the specificity of the Antibody and One-Day ChIP Kit combination.
