Sonicated chromatin from HeLa cells (20 µg) was immunoprecipitated with 2 µg of anti-H3K4me1 antibody (blue bars) or control IgG (red bars) for 2 h using the ChampionChIP One-Day Kit (GA-101). The resulting input and ChIP DNA fractions were characterized with qPCR primers specific for the regions one, two, and four kilobases downstream of the CDKN1A transcription start site (TSS, +1 kb, +2 kb, and +4 kb Tiles) to calculate the percentage of co-precipitating DNA relative to input. The immunoprecipitation using anti-H3K4me1 enriched genomic DNA at least four kb downstream of the TSS with a high signal-to-noise ratio relative to the control IgG immunoprecipitation, demonstrating the efficiency of the Antibody and One-Day ChIP Kit combination. In contrast, the immunoprecipitation less significantly enriched genomic DNA within 2 kb of the TSS, demonstrating the specificity of the Antibody and One-Day ChIP Kit combination. Back to Top
