How It Works: Speed and Ease
Immunoprecipitation Done in 2 Hours with ChampionChIP Antibody Using
an optimized IP buffer system provided by the ChampionChIP One-Day Kit, the
efficiency and specificity of the ChIP-grade antibody are maximized such that
immunoprecicipation can be done within 2 hours. We guaranteed high
signal-to-noise ratio and low background when the Champion Antibody kit is used
in combination of ChampionChIP One-Day Kit. In addition, these antibody kits are
the first ones in the market that include positive and negative control qPCR
primers, insuring the accuracy of ChIP analysis. The figure depicts the workflow
of the entire ChampionChIP System including One-Day ChIP Kit, ChIP-Grade
Antibody Kit, and FREE Excel-based data analysis template.

Sonicated chromatin from HeLa cells (20 µg) was immunoprecipitated with 2 µg of anti-H3K9me2 antibody (blue bars) or control IgG (red bars) for 2 h using the ChampionChIP One-Day Kit (GA-101). The resulting input and ChIP DNA fractions were characterized with qPCR primers specific for the GAPDH, MYOD1 and SAT2 genomic loci to calculate the percentage of co-precipitating DNA relative to input. The immunoprecipitation using anti-H3K9me2 enriched genomic DNA representing heterochromatin (the SAT2 locus) with a high signal-to-noise ratio, demonstrating the efficiency of the Antibody and One-Day ChIP Kit combination. In contrast, the immunoprecipitation less significantly enrich genomic DNA representing transciptionally inactive euchromatin (the MYOD1 locus) and did not enrich genomic DNA representing actively transcribed euchromatin (the GAPDH locus) relative to the immunoprecipitation using control IgG, demonstrating the specificity of the Antibody and One-Day ChIP Kit combination.

Shelf Life
