Internal Normalizers
The GAPD Internal Normalizer allows researchers without access to real-time
equipment to perform quantitative RT PCR analyses. This unique primer mix is
designed to detect the expression level of GAPD, a commonly used
housekeeping gene, in the same tube as your gene of interest. Just add the
Internal Normalizer to a standard PCR containing your first strand cDNA
template (synthesized from total RNA) and PCR primers specific for your gene
of interest.
The special Internal Normalizer primer mix attenuates the GAPD signal
placing it in the same dynamic range as many other genes. The resulting GAPD
signal does not saturate nor interferes with the detection of the gene of
interest. The expression level of the gene of interest is calculated as the
target-to-Normalizer signal intensity ratio. These ratios are then compared
between experiments to obtain relative expression values. This special
primer mix serves as an effective internal control for systematic errors
such as pipet error in template loading resulting in more accurate relative
gene expression results.
