SureSilencing™ siRNA Array Layout and Controls

Figure: The Well-Designed Layout of the SureSilencing siRNA Array

Gene-Specific siRNA
Wells A1 through G12 contain duplicate wells of siRNA pairs targeting a thoroughly researched panel of 42 relevant, pathway-focused genes. Immediately adjacent wells contain the replicates for easy analysis of the phenotypic assay data readout.

Negative Control for RNA Interference
Wells H1 through H4 contain a negative control siRNA (NEG). Compare or normalize the results of your phenotypic assay to this negative control to ascertain the gene-specific siRNA effects on the assay.

Phenotypic Assay Normalization
Wells H5 through H8 do not contain siRNA (NOR) but are plated with your cells to serve as a mock transfection. Use a biological treatment (control or experimental) on these wells that produces a reliable and detectable signal from your phenotypic assay. Using this signal to normalize the raw signals controls for plate-to-plate variation.

Transfection Efficiency Control
Wells H9 and 10 contain the fluorescently labeled negative control siRNA and serve as a transfection efficiency control (TEC) when monitored by fluorescence microscopy.

Phenotypic Assay Background
Wells H11 and 12 do not contain siRNA and will not be plated with cells (NOC) to serve as a measure of the background in your phenotypic assay.
